The study was conducted on a national Danish sample of 1,983,785 people aged 60 to 75 years between 2000 and 2018 and included only people who had not previously been diagnosed with dementia or treated with specific dementia medications.

During the study, 99,384 people developed dementia, and 469,920 people were used as the control group. PPI use has been associated with an increased risk of all-cause dementia, especially in people aged 60 to 69 years at diagnosis. In the 60 to 69 year group, incidence ratios ranged from 1.25 to 1.59 depending on duration of use and 1.36 overall for ever PPI users.

A less negligible risk was observed in the 70 to 79 and 80 to 89 age groups: 1.12 and 1.06 for chronic use, respectively. No significant association was observed between PPI use and dementia in people over 89 years of age at diagnosis.
